Pada proses pembuatan yogurt, terjadi konversi gula susu (laktosa) menjadi asam laktat melaluiproses fermentasi oleh bakteri asam laktat (BAL). Yogurt umumnya dibuat dengan menambahkankultur bakteri yang mengandung 2 jenis bakteri asam laktat, yaitu Lactobacillus bulgaricus (L. bulgaricus) dan Streptococcus thermophilus (S. thermophillus). Pada penelitian ini dilakukanpenambahan BAL lain, yaitu Lactobacillus acidophilus (L. acidophilus) untuk membandingkankualitas yogurt yang dihasilkan. Variasi perbandingan konsentrasi bakteri yang digunakan adalah1:1, 1:4 dan 4:1 untuk kultur dua bakteri (L. bulgaricus : S. thermophilus) dan 1:1:1, 1:4:1 dan4:1:1 untuk kultur tiga bakteri (L. bulgaricus : S. thermophilus : L. acidophilus). Susu murni di pasteurisasi pada suhu 85°C selama 15 menit dan inkubasi dilakukan selama 8 dan 10 jam padasuhu 42°C. Parameter yang diuji diantaranya adalah pH, kadar asam laktat, kadar protein dankadar laktosa. Hasil penelitian menunjukkan bahwa penambahan L. acidophilus dengan perbandingan (L. bulgaricus : S. thermophilus : L. acidophilus = 1:4:1) memberikan hasil terbaikpada jam ke 10 dengan pH terendah sebesar 4,56, kadar asam laktat sebesar 1,12% (b/v), kadarprotein 0,30% (b/v) dan kadar laktosa 1,09% (b/v).

In yogurt manufacture process. lactose is converted to lactic acid through fermentation by lactic acid bacteria (LAB). Yogurt is usually made by adding bacterial culture that contains Iwo bacteria. which are Lactobacillus bulgaricus (L. bulgaricus) and Streptococcus thermophilis (S. thermophilus). the present study, we add one more bacteria. which is Lactobacilus acidophilus (L. acidophilus) to compare the quality of the yogurts. The ratio of bacteria used in the present study was 1:1. 1:4 and 4:1 for two bacterial culture (L. bulgaricus : S thermophilus). and 1:1:1. 1:4:1. 4:1:1 for three bacterial culture (. bulgaricus : S. thermophilus : L. acidophilus). Cow milk was pasteurized at 85°C for 15 minutes and incubated for 8 and 10 hours at 42°C. The determined parameters were pH. lactic acid content. protein content and lactose content. The results of the present study showed that addition of L. acidophilus at 1:4:1 ratio (L. bulgaricus : S. thermophilus : L acidophilus) gave the best result at 10 h incubation with pH value of 4.56. lactic acid content of 1.12% (w/v). protein content of 0.30% (w/v) and lactose content of 1.09% (w/v).
