Tongue cancer is a disease that often unpredictable, aggressive, with poor prognosis, which require a fast. accurate, and effective treatment with minimum side effects. Herbal medicine is the treatment with promising prospective thus leading to the search of bioactive compound with anticancer potential. Mangosteen (Garcinia mangostana Linn.) with its xanthone derivative compound content is a potential candidate of herbal medicine. This study is aimed at identification of xanthone derivative compound from mangosteen pericarp with anticancer potential and tests them through the suppression of VEGF expression in SP-C1 tongue cancer cell lines. This study was a laboratory experimental study using SP-C1 tongue cancer cell lines. Vacuum liquid chromatography. column chromatography. and preparative thin layer chromatography methods were used to isolate the xanthone derivative compound from ethanol extract of mangosteen pericarp followed by compound structure identification with NMR 1H and 13C spectroscopy. Cytotoxicity test using MTT assay was performed to get compounds with anticancer potentials. Then were tested for VEGF expression analysis. Data analysis were then tested using ANAVA. with a level of significance α = 0.05. The results revealed nine compound identified as xanthone derivative. which include three most studied xanthoncs: ϓ-mangosteen. α-mangosteen. and β-mangosteen. The active compounds that act as anticancer are a-mangostin. 8-hydroxicudraxanthone G. Cudraxanthone G. Garsinone D. and Garsinone C. Overall. xanthone derivative compounds have the potentials to suppress VEGF expression (F cale =3.2. ξ/K = 0.100). It can he concluded that xanthone derivative compound has the potentials as the candidate of herbal medicine with the activity to suppress VEGF expression.
