Gugus pelindung yang labil terhadap cahaya (PPGs) adalah suatu alat kunci untuk menginvestigasi proses biokimia dalam sel hidup akibat dari kemampuannya untuk melepaskan suatu substrat tanpa dibatasi ruang dan waktu. 1,2 Gugus p-Hidroksifenasil telah terbukti menjadi gugus pelindung yang labil terhadap cahaya (PPG) yang sangat efisien untuk digunakan terhadap macam-macam substrat seperti, halida, fenol, dan asam. Mekanisme pelepasan gugus pelindungnya dengan cahaya melibatkan suatu proses Intersystem crossing (ISC) yang sangat cepat menjadi suatu triplet biradikal, diikuti dengan pelepasan substrat secara bertahap dan pembentukan serentak suatu triplet biradikal yang dekat dengan senyawa antara spirodion. Hidrolisis senyawa antara tersebut menghasilkan asam p-hidroksifenilasetat. 3 Pelepasan gugus pergi yang baik seperti dietil fosfat, tosilat dan mesilat berlangsung sangat efisien dengan quantum yield (~1) dan kecepatan pelepasannya yang sangat cepat sekali (k ~ 10 10 s-1). Akan tetapi, quantum yield menjadi turun secara signifikan untuk gugus pergi yang jelek (~ 0.05 untuk fenol). Suatu proses tautomerisasi adiabatik4 kemungkinan besar terlibat secaran kompetitif dengan proses pelepasan gugus pergi. Proses samping yang membuang energi ini berakibat pada menurunnya quantum yield PPG pHP. Pada penelitian yang berkelanjutan ini, kita telah mensintesis p-Hidroksifenasil fenilasetat (pHP-PA) dan mempelajari bagaimana pengaruh konsentrasi suatu basa umum dan pH terhadap quantum yield pelepasan gugus pelindung pHP-PA secara keseluruhan.

Photoremovable protecting groups (PPGs) are a key tool for investigation of biochemical processes in living cells due to their ability to release the substrates in time and space of intererest. 1,2 p-Hydroxyphenacyl moiety has been proved to be an efficient photoremovable protecting group (PPG) for various substrates, for example, halides, phenols and acids. The mechanism of their photodeprotection involves a rapid ISC to the triplet state followed by a subsequent release of the substrate and concomitant formation of a triplet biradical that closes to the putative spirodione intermediate. Hydrolysis of the putative spirodione intermediate formed p-hydroxyphenylacetic acid.3 The release of good leaving groups such as diethyl phosphate, tosylate and mesylate, was efficient with high quantum yields (~1) and ultrafast release rates (k ~ 10 10 s-1). However, the quantum yield drops significantly for poor leaving group (~ 0.05 for phenol). An adiabatic triplet state tautomerization4 might represent the energy wasting paths decreasing the quantum yield of the pHP PPG. In the present work, we synthesized p-Hydroxyphenacyl phenylacetate (pHP-PA)and studied how the concentration of a general base and pH affects the overall disappearance quantum yields of pHP-PA.
