Pengaruh Modifikasi Kimiawi

Kemampuan untuk menghasilkan enzim yang stabil sangatlah penting untuk pemakaiannya sebagai biokatalis. Tujuan penelitian ini adalah untuk meningkatkan kestabilan α-amilase Saccharomycopsis fibuligera melalui berbagai modifikasi kimiawi secara selektif terhadap rantai samping lisina pada permukaan enzim. Modifikasi dilakukan dengan hidrofilisasi dengan asam glioksilat, pembentukan ikatan silang dengan dimetiladipimidat, serta peningkatan interaksi hidrofobik dengan anhidrida asam asetat. Enzim yang dimodifikasi dengan anhidrida asam asetat menunjukkan peningkatan faktor stabilisasi yang paling tinggi yaitu 11,9 kali, juga peningkatan afinitas terhadap substrat amilosa sebesar 32%, peningkatan konstanta spesifisitas sebesar 23,8%, bila dibandingkan dengan α-amilase sebelum modifikasi.

The ability to obtain stable enzymes is crucial for their application as biocatalysts. The objective of this study was to increase the stability of Saccharomycopsis fibuligera α-amylase by various selective chemical modifications of lysine side chains on the enzyme surface. Modifications were performed by hydrophilization with glyoxylic acid, by cross-link formation with dimethyladipimidate, by increasing hydrophobic interactions with acetic acid anhydride. In the comparison with the unmodified α-amylase, acetic acid anhydride modified enzyme showed the highest stabilization factor i.e. 11.9 fold, an increase of the affinity toward amylose substrate i.e. 32%, also an increase of the specificity constant i.e. 23.8%.
