Alteration of Leaf Anatomy Structure in Mangosteen Regenerants In Vitro Caused by Gamma Ray Irradiation

Mangosteen has a low of rate photosynthesis. Research was conducted to study the alteration of leaf structure of mangosteen caused by gamma ray irradiation. Leaf samples from 21 mutant regenerants and from a selected control were examined using both paradermal and transverse sections. Paradermal sectioning was conducted by using a whole mount method followed by staining with safranin 1%, whereas transverse sections were prepared using paraffin methods where the leaf was sliced sing a rotary microtome at 10 pm thickness and then stained with saffranin 1% and hstgreen 0.5%. Gamma ray irradiation altered leaf structure. Several mutant regenerants had a higher stomata density and a higher stomatal index than control regenerants-those regenerants were R-5/2, R-5/3, R-5/4, R-10/4, R-15/3, R-2511, R-30/1 and R-30/2. Transverse sections showed that the thickness of the upper cuticle of mutant regenerants was thinner than control regenerants, except for the mutant regenerant R-5/1. Regenerants R-1014, R-15/1, R-15/2, R-15/3 had a thicker palisade parenchyma, spongy mesophyll and leaf lamina than control regenerants. In addition, tie spongy mesophyll was thicker and the number of vascular bundles was also greater than in the control regenerant.
