This research was conducted to find out the effect of sappan wood extract on blood antioxidant profile of mice based on the measurement of malondialdehida (MDA) level, superoxide dismutase (SOD) activity, glutathione peroxside (GPx) level, and total antioxidant status (SAT). Sappan wood extract of 200 mg/kg bw, 400 mg/kg bw, 800 mg/kg bw, vitamin E 60 mg/kg bw, dan vitamin C 60 mg/kg bw were given to mice everyday for 15 days. Aflatoxin with total dose of 375 pg/kg bw dan 750 pg/kg bw were given to mice on the first and eighth days. Aflatoxin was given one hour after antioxidant administration. At the sixteenth, mice were killed and blood was taken from the heart. To separate between blood trombocyte and plasma, blood was deposited at room temperature or at refrigerator. After separation, blood plasma was kept in vial and used. to measure MDA levels, SOD activities, GPx levels, dan total antioxidant defense profiles. The results showed that aflatoxin decrease the antioxidant defense system. Furthermore, added supplement antioxidant increased the antioxidant defense system Sappan wood extract was relatively effective compared to vitamin E and vitamin C. Sappan wood extract of 400 mg/kg bw is the most effective antioxidant to increase blood antioxidant system on mice exposured to aflatoxin at low dose as well as at high dose compared to sappan wood extract of 200 or 800 mg/kg bw.
